Product Description:

Tafenoquine is an 8-aminoquinoline antimalarial, related to primaquine but with a much longer half-life of around two weeks. That single property is what makes it significant: it kills the dormant liver forms of Plasmodium vivax that cause relapses months after the original infection, and it does so in a single dose where primaquine requires fourteen days of treatment that patients frequently do not complete.

It is used for radical cure of P. vivax malaria, given with a blood-stage drug such as chloroquine, and at a different dose for prophylaxis in travellers, taken weekly. The constraint on its use is glucose-6-phosphate dehydrogenase deficiency: in people with the enzyme deficiency, 8-aminoquinolines cause acute haemolysis, and because tafenoquine cannot be stopped once taken, quantitative G6PD testing is required before every course — not the qualitative test that sufficed for primaquine. It is contraindicated in pregnancy and breastfeeding where the infant's status is unknown.
