Product Description:

This tablet pairs atovaquone with proguanil, and the combination is synergistic rather than merely additive. Atovaquone collapses the parasite's mitochondrial membrane potential by blocking cytochrome bc1; proguanil, through a mechanism separate from its usual antifolate action, lowers the concentration of atovaquone needed to do so. The pairing also suppresses the emergence of resistance that atovaquone alone develops quickly.

It is used to prevent and treat uncomplicated falciparum malaria. As prophylaxis it is popular with travellers because it is started only one to two days before entering a malarious area and stopped seven days after leaving, unlike older drugs needing weeks of continuation, and because it is well tolerated. It acts against liver-stage parasites, which is what allows the short post-travel course, but it does not clear the dormant liver forms of vivax malaria. Absorption depends on food or a milky drink. It is avoided in severe renal impairment. The paediatric tablet is a quarter strength.
