Product Description:

Crizotinib was the first ALK inhibitor to reach the clinic, and it established the principle that a lung cancer defined by a genetic rearrangement can be treated as a distinct disease. It inhibits ALK, ROS1 and MET. In tumours carrying an ALK or ROS1 fusion gene the rearranged kinase is constitutively active and the cell depends on it entirely, so blocking it produces rapid, often dramatic responses in patients who would otherwise face conventional chemotherapy.

It is used for ALK-positive or ROS1-positive advanced non-small cell lung cancer and for ALK-positive anaplastic large cell lymphoma and inflammatory myofibroblastic tumour in younger patients. Visual disturbances — trails and flashes on changing light levels — are reported by most patients and are usually mild. Nausea, oedema, transaminase elevation, QT prolongation and bradycardia occur. Resistance emerges within a year or two, typically through new ALK mutations or brain metastases, since central nervous system penetration is limited.
