Ixazomib is used in adults to treat multiple myeloma (bone marrow cancer), in combination chemotherapy with lenalidomide and dexamethasone. This combination is usually given after treatment with at least one other cancer medicine has failed.

Product Description:
Ixazomib inhibits the proteasome, the cell's protein disposal machinery. Myeloma cells produce immunoglobulin in enormous quantities and depend on that machinery more heavily than any normal cell, so blocking it causes misfolded protein to accumulate to lethal levels. What separates ixazomib from earlier proteasome inhibitors is that it is absorbed orally and binds reversibly with a short dissociation time, allowing weekly rather than twice-weekly dosing.
It is used for multiple myeloma in combination with lenalidomide and dexamethasone, in patients who have received at least one prior therapy, taken once weekly for three weeks of a four-week cycle. Because all three drugs in the regimen are oral, the whole treatment can be delivered without infusion visits, which changes what is feasible for patients distant from a centre. Thrombocytopenia follows a predictable cyclical pattern; diarrhoea, constipation, nausea, rash and peripheral neuropathy occur, the neuropathy notably less than with injectable agents.
