Delafloxacin is a broad-spectrum fluoroquinolone antibiotic used to treat acute bacterial skin and skin structure infections (ABSSSI) and community-acquired bacterial pneumonia (CABP). It works by inhibiting bacterial enzymes DNA gyrase and topoisomerase IV, which are essential for DNA replication and cell division, leading to bacterial death. Delafloxacin is active against both Gram-positive (including MRSA) and Gram-negative bacteria.

Product Description:

Delafloxacin is a fluoroquinolone with an unusual property: it is anionic, where other quinolones are zwitterionic. That charge gives it enhanced potency in acidic environments — abscesses, inflamed skin, infected tissue — where conventional quinolones lose activity. It is also active against methicillin-resistant Staphylococcus aureus, which most fluoroquinolones are not.

It is indicated for acute bacterial skin and skin structure infection and for community-acquired bacterial pneumonia, and is supplied as 450 mg tablets and as powder for injection, with the oral and intravenous doses designed for direct switch.

The fluoroquinolone class warnings apply in full. Regulators including the FDA and EMA have restricted the class following reports of disabling and potentially irreversible tendon, muscle, joint and nervous system effects, together with aortic aneurysm risk. Fluoroquinolones are no longer recommended for uncomplicated infections where alternatives exist. Labelling must carry the current class warnings for the destination market.

Sourcing notes. Delafloxacin is a relatively recent product and patent position varies by territory; verify freedom to operate in the destination market before treating it as genericisable.
