Danazol is a synthetic androgen (male hormone derivative) used to treat endometriosis, fibrocystic breast disease, and hereditary angioedema. It works by suppressing pituitary gonadotropin secretion, leading to reduced estrogen production. This helps relieve pain and symptoms associated with hormone-dependent conditions.

Product Description:

Danazol is a synthetic derivative of ethisterone with a mixture of actions: it suppresses the pituitary release of LH and FSH, binds androgen and progesterone receptors, and displaces testosterone from sex hormone binding globulin, raising free testosterone. The result is a low-oestrogen, high-androgen state in which endometrial tissue, inside and outside the uterus, becomes atrophic.

It is used for endometriosis, for fibrocystic breast disease and menorrhagia in some markets, and — through a quite separate mechanism — for hereditary angioedema, where it increases hepatic production of C1 esterase inhibitor and prevents attacks. Its androgenic effects limit gynaecological use and are why it has largely been replaced by GnRH analogues and progestins for endometriosis: weight gain, acne, oily skin, hirsutism, and voice deepening that may not reverse. It is contraindicated in pregnancy because it can virilise a female fetus, so non-hormonal contraception is required, and it raises liver enzymes and lowers HDL cholesterol.
