Cefaclor is a second-generation cephalosporin antibiotic that works by inhibiting bacterial cell wall synthesis, leading to bacterial cell death. It is effective against a wide range of Gram-positive and some Gram-negative bacteria. Commonly prescribed for infections like otitis media, pneumonia, bronchitis, urinary tract infections, and skin infections.
Product Description:
Cefaclor is a second-generation oral cephalosporin with improved Gram-negative activity over first-generation agents while retaining useful Gram-positive cover. Its established uses are otitis media, pharyngitis, and lower respiratory and urinary tract infections.
Two characteristics shape its use. It is less stable to beta-lactamases than later oral cephalosporins such as cefuroxime axetil, which has reduced its role as resistance has spread. And it carries a recognised association with serum-sickness-like reactions, particularly in children — an immune-mediated response with rash, arthralgia and fever appearing days into treatment, more frequently reported with cefaclor than with other cephalosporins.
It is supplied as capsules and tablets at 250 and 500 mg, an extended-release 750 mg tablet, and oral suspension at 125 and 250 mg per 5 mL.
Sourcing notes. The suspension carries most paediatric volume and requires liquid orals capability with stability data matched to the destination climatic zone — Zone IVb data is frequently the binding constraint for South Asian, Southeast Asian and African markets. WHO classifies cefaclor under Watch, which affects its position in stewardship-driven procurement. The extended-release tablet is a separate formulation requiring its own dissolution validation and should not be quoted interchangeably with immediate-release.
