Product Description:

Ribociclib inhibits cyclin-dependent kinases 4 and 6, arresting the cell cycle before DNA replication in tumours whose growth depends on that machinery. Among the three drugs in its class it is the one with the clearest overall survival benefit in hormone receptor-positive advanced breast cancer, demonstrated across trial populations including premenopausal women — a group historically under-represented in breast cancer trials.

It is used with an aromatase inhibitor or fulvestrant in hormone receptor-positive, HER2-negative advanced breast cancer, and in high-risk early breast cancer after surgery, taken for three weeks of each four-week cycle. Neutropenia is common and reflects reversible marrow cell cycle arrest rather than cell destruction. Two class-atypical effects require monitoring: QT interval prolongation, so an ECG is taken at baseline and during the first cycles and interacting drugs are reviewed, and hepatotoxicity, requiring liver function testing on a defined schedule.
