Propylene glycol (IUPAC name: propane-1,2-diol) is a colorless, viscous liquid with a faintly sweet taste and almost no odor. Chemically represented as CH3CH(OH)CH2OH, it belongs to the class of diols, containing two hydroxyl (–OH) groups, which gives it excellent solubility and versatility. As an aliphatic glycol, propylene glycol is miscible with a wide range of solvents, including water, acetone, and chloroform, making it a highly adaptable excipient in pharmaceutical, cosmetic, and food formulations.

In pharmaceuticals, propylene glycol is widely used as a solvent, humectant, and carrier for oral, topical, and parenteral formulations. Why DEG and EG testing matters

Diethylene glycol (DEG) and ethylene glycol (EG) are toxic industrial solvents. Their presence in propylene glycol or glycerin, through contamination or deliberate substitution, has caused repeated mass poisonings, including the deaths of children in The Gambia and Uzbekistan in 2022, which WHO linked to contaminated cough syrups made by Indian manufacturers. USP limits each of DEG and EG in propylene glycol to not more than 0.10%, and Ph. Eur. has required a DEG and EG test since January 2025. India has required government-lab testing of cough syrups for export since June 2023, and after child deaths linked to Coldrif syrup in 2025 its drug regulator ordered strict enforcement of batch testing of excipients.
