Metaxalone is a centrally acting skeletal muscle relaxant. Its mechanism has never been established precisely; it is thought to act by general depression of the central nervous system rather than by any direct effect on muscle, on the motor end plate or on the nerve fibre. Its practical appeal is that it causes noticeably less sedation than methocarbamol or cyclobenzaprine, which matters for patients who must function during the day.

It is used for short-term relief of discomfort from acute, painful musculoskeletal conditions, alongside rest and physiotherapy. Food substantially increases absorption, so whether it is taken with meals affects the dose that actually reaches the patient. It is contraindicated in significant liver or kidney impairment and in patients with a history of drug-induced haemolytic anaemia. It has weak monoamine oxidase inhibitory activity, and serotonin syndrome has been reported when it is combined with SSRIs, SNRIs and other serotonergic drugs.
