Dapsone is a sulfone-class antibiotic primarily used to treat leprosy (Hansen’s disease), dermatitis herpetiformis, and certain bacterial skin infections. It works by inhibiting bacterial synthesis of dihydrofolic acid, slowing bacterial growth. Dapsone also has anti-inflammatory properties, making it effective in managing chronic dermatological conditions. It is available in oral and topical forms for systemic and local therapy.
Product Description:
Dapsone is a sulfone antibacterial, inhibiting folate synthesis in a manner similar to the sulfonamides. Its two principal roles are quite separate. It is a cornerstone of multidrug therapy for leprosy, used with rifampicin and clofazimine in the WHO regimens. And it treats dermatitis herpetiformis, where its effect appears to be anti-inflammatory rather than antibacterial. It is also used for Pneumocystis jirovecii prophylaxis where co-trimoxazole is not tolerated.
It is supplied as 25, 50 and 100 mg tablets, and as a topical gel for acne.
Two haematological cautions govern its use. Dapsone causes dose-related haemolysis and methaemoglobinaemia, markedly more severe in glucose-6-phosphate dehydrogenase deficiency — G6PD screening before treatment is standard where the deficiency is prevalent, which includes much of Africa, the Mediterranean and South and Southeast Asia. Dapsone hypersensitivity syndrome, appearing weeks into treatment with fever, rash and hepatitis, is rare but can be fatal.
