Product Description:

Crisaborole is a boron-containing small molecule that inhibits phosphodiesterase 4. PDE4 breaks down cyclic AMP inside inflammatory cells; inhibiting it raises cyclic AMP, which suppresses production of the cytokines that drive atopic dermatitis. The boron atom is what allows a molecule this small to bind the enzyme and to penetrate skin, and the drug is rapidly metabolised once absorbed, so systemic exposure stays low.

It is used twice daily for mild to moderate atopic dermatitis in patients aged three months and over. Its value is that it is not a steroid: it can be used on the face, eyelids and flexures, and for long periods, without the skin thinning that limits topical steroids — which matters in a chronic relapsing disease that often starts in infancy. Its effect is modest compared with a potent steroid. Application site pain — burning or stinging — is common, particularly in the first days and on inflamed skin, and is the usual reason patients stop.
