Product Description:

Suzetrigine is a first-in-class, selective inhibitor of NaV1.8, a voltage-gated sodium channel found almost exclusively in peripheral pain-sensing neurons. Blocking NaV1.8 stops pain signals at their source, before they reach the spinal cord, and because the channel is not present in the brain, the drug does not cause the sedation, euphoria or respiratory depression that come with opioids. It was the first new mechanism for acute pain approved in more than two decades.

It was approved by the FDA in January 2025 for moderate to severe acute pain in adults, on the basis of trials after abdominoplasty and bunionectomy. It was better than placebo, but in those trials not better than hydrocodone with paracetamol, so its advantage is safety and absence of addiction potential rather than stronger pain relief. A loading dose is followed by maintenance doses every twelve hours. Strong CYP3A inhibitors are contraindicated, and it can reduce the effectiveness of hormonal contraceptives.
