Podophyllotoxin (PPT) is the active pharmaceutical ingredient in Podofilox, a topical medication widely used for the treatment of external genital warts associated with human papillomavirus (HPV) and molluscum contagiosum infections. It is intended only for visible external lesions and is not recommended for internal HPV infections without warts. The medication can be applied either by a healthcare professional or by patients themselves, following prescribed instructions.
Podophyllotoxin is a non-alkaloid lignan derived from the roots and rhizomes of plants belonging to the Podophyllum genus. It works as a potent antimitotic agent by inhibiting cellular and viral DNA replication, disrupting microtubule formation, and preventing cell division, which leads to controlled destruction of wart tissue. Due to its targeted topical action, it is considered safer than crude podophyllum resin, which is associated with higher toxicity and adverse effects.
Podophyllotoxin is commonly formulated as a 0.5% cream, gel, or solution. The standard treatment regimen involves twice-daily application for three consecutive days, followed by four days without application, repeated for up to four weeks.
