Methoxsalen, also known as Xanthotoxin or 8-methoxypsoralen, is a photosensitizing medication primarily used in combination with ultraviolet A (UVA) light therapy to treat a range of skin conditions, including psoriasis, eczema, vitiligo, and certain cutaneous lymphomas. Marketed under brand names such as Oxsoralen and Melanocyl, methoxsalen works by modifying the way skin cells respond to UVA radiation, helping to reduce abnormal cell proliferation and restore pigmentation in affected areas.
Methoxsalen belongs to the furanocoumarin class of organic compounds, which are naturally occurring molecules composed of coumarin fused with a furan ring. Its unique chemical structure allows it to intercalate into DNA and, upon activation by UVA light, form cross-links that slow the growth of overactive skin cells. The therapy, commonly known as PUVA (psoralen + UVA), is carefully dosed based on individual patient sensitivity, historically assessed using the Fitzpatrick skin type scale to minimize phototoxicity.
