Product Description:
Linaclotide is a fourteen-amino-acid peptide agonist of guanylate cyclase-C on the luminal surface of intestinal epithelium. Activation raises cyclic GMP inside the cells, which opens the CFTR chloride channel and draws chloride, bicarbonate and water into the gut lumen, speeding transit and softening stool. Cyclic GMP released from the cells also reduces the firing of pain-sensing nerves, so it relieves abdominal pain as well as constipation. It acts locally and is barely absorbed.
It is used for irritable bowel syndrome with constipation and chronic idiopathic constipation in adults, and in the US for functional constipation in children aged six and over. It is taken once daily at least thirty minutes before the first meal. Diarrhoea is the main adverse effect and the main reason patients stop. It is contraindicated in children under two because of dehydration deaths in juvenile animal studies.
