Dacarbazine is used in the treatment of cancers of lymphatic system and malignant melanoma (a type of skin cancer). It may be also used to treat other conditions, as determined by the doctor.

Product Description:

Dacarbazine is another prodrug, activated in the liver to a methylating species that transfers a methyl group to DNA guanine. The resulting lesion is repaired by the enzyme MGMT, and tumours expressing high levels of that enzyme resist the drug — the same mechanism that governs temozolomide, which is essentially an orally bioavailable relative delivering the same active fragment.

It is used for metastatic malignant melanoma, where it was the standard of care for decades before immune checkpoint inhibitors and BRAF-targeted therapy displaced it, and it remains a component of ABVD for Hodgkin lymphoma and of regimens for soft tissue sarcoma. Nausea and vomiting are severe and were historically its limiting feature. Myelosuppression, a flu-like syndrome, photosensitivity and painful extravasation occur; hepatic vein thrombosis with hepatocellular necrosis is a rare but fatal complication.

Sourcing notes. Off patent, supplied as a lyophilised powder for reconstitution. The solution is light-sensitive and degrades quickly, so infusion lines are shielded and the product must be protected from light throughout storage and transport. Cytotoxic handling. Demand persists mainly through Hodgkin lymphoma protocols rather than melanoma.
