Product Description:
Amisulpride is a substituted benzamide that blocks dopamine D2 and D3 receptors, with an unusual dose-dependent profile. At low doses it preferentially blocks presynaptic autoreceptors, which increases dopamine release and improves negative symptoms such as withdrawal and flat affect; at higher doses it blocks postsynaptic receptors and treats positive symptoms such as hallucinations and delusions. It is highly selective, with almost no activity at histamine, muscarinic or adrenergic receptors, so it causes little sedation or weight gain compared with olanzapine.
Oral amisulpride is used for schizophrenia. Separately, a low-dose intravenous formulation is approved in the United States for preventing and treating post-operative nausea and vomiting, at doses far below antipsychotic ones — the same molecule in an entirely different role. Raised prolactin is the characteristic adverse effect, causing menstrual disturbance, galactorrhoea and sexual dysfunction. It prolongs the QT interval, and is cleared by the kidneys, so the dose is reduced in renal impairment.
Sourcing notes. The oral tablet and the intravenous anti-emetic are separate products with separate registrations and completely different buyers. Buyers should state which is required. Oral amisulpride is registered in Europe and much of Asia but not the US.
