Product Description:

Prucalopride is a highly selective serotonin 5-HT4 receptor agonist. Stimulating 5-HT4 receptors on enteric neurons increases acetylcholine release, strengthening the high-amplitude propagating contractions that drive colonic content forward. That action makes it a true prokinetic rather than a stool softener or secretagogue, which is useful when constipation reflects slow transit and laxatives have failed.

It is used for chronic idiopathic constipation in adults whose symptoms have not responded to laxatives. Earlier 5-HT4 drugs, cisapride and tegaserod, were withdrawn or restricted because of cardiovascular problems linked to activity at other receptors, including the hERG potassium channel. Prucalopride's selectivity avoids that, and a large observational study found no increase in cardiovascular events. Headache, nausea, diarrhoea and abdominal pain are common, mostly on the first day. The dose is reduced in severe renal impairment and in older patients in some markets.
