Product Description:

This tablet combines two nucleoside reverse transcriptase inhibitors, lamivudine 150 mg and zidovudine 300 mg. Both are taken up into cells, converted to triphosphates and built into viral DNA, where they stop the chain and block HIV replication. Zidovudine was the first antiretroviral, and this pairing served as a standard NRTI backbone for many years. The film-coated tablet gives a full adult dose of both drugs, taken twice daily, always with a third antiretroviral.

WHO guidelines have moved first-line treatment to dolutegravir with tenofovir and lamivudine or emtricitabine, so this backbone is now used mostly in second-line regimens after tenofovir failure, and in some children. It is taken with or without food. Zidovudine suppresses bone marrow, so haemoglobin and neutrophil counts are checked, and severe anaemia or neutropenia rules it out. Nausea, headache, nail pigmentation, lipoatrophy and lactic acidosis can occur. Stavudine must not be combined with it. Stopping lamivudine in hepatitis B co-infection can trigger a hepatitis flare.
