Product Description:
Paclitaxel is a taxane that stabilises microtubules and so freezes cancer cells in mitosis. In liposomal paclitaxel the drug is carried in phospholipid vesicles rather than dissolved in polyoxyl castor oil and ethanol, the solvent of conventional paclitaxel that is linked to hypersensitivity and needs non-PVC infusion sets. It is also distinct from the albumin-bound nanoparticle form. Liposomal products are generally freeze-dried powders in vials, so confirm the vial content and the reconstitution volume.
Where it is registered, liposomal paclitaxel is used in ovarian, breast and non-small cell lung cancer, often with a platinum drug. The three paclitaxel formulations differ in dose, infusion time and premedication, so they cannot be swapped milligram for milligram, and each must be prescribed and labelled by its full name. Neutropenia, peripheral neuropathy, hair loss and hypersensitivity remain the main risks, and blood counts are checked before each cycle. Paclitaxel is metabolised by CYP2C8 and CYP3A4, so strong inhibitors or inducers of these enzymes need review.
Sourcing notes. Liposomal paclitaxel is registered only in some markets, so confirm local status before quoting it against a conventional paclitaxel tender. The specification should cover the liposome itself, particle size, encapsulation efficiency and lipid content, as well as assay, related substances, sterility and endotoxin. Liposomal forms are often more temperature-sensitive than solutions, so read the label for storage and cold chain needs. Request a lot-specific CoA, the GMP certificate and a CoPP. It is a cytotoxic, prescription-only medicine; name the formulation explicitly in your enquiry.
