Indomethacin is a synthetic nonsteroidal indole derivative with potent anti-inflammatory and analgesic properties. As a nonsteroidal anti-inflammatory drug (NSAID), it exerts its therapeutic effects primarily through inhibition of the cyclooxygenase (COX) enzymes, thereby reducing the synthesis of prostaglandins responsible for pain, inflammation, and fever. It is widely used in the management of inflammatory conditions such as rheumatoid arthritis, osteoarthritis, ankylosing spondylitis, acute gout, and other musculoskeletal disorders.
Beyond its established anti-inflammatory activity, indomethacin has demonstrated potential chemopreventive and anticancer-related properties in experimental settings. By inhibiting cyclooxygenase-mediated processes, it may reduce DNA adduct formation caused by heterocyclic aromatic amines. Additionally, indomethacin has been observed to modulate cellular mechanisms involved in tumor progression, including inhibition of multidrug-resistant protein type 1 (MDR1), which may enhance the efficacy of certain antineoplastic agents in resistant tumors.
