Product Description:

Dactinomycin, also called actinomycin D, is an antibiotic isolated from Streptomyces that turned out to be far too toxic for infection and precisely useful in cancer. It intercalates between guanine and cytosine base pairs in DNA and blocks RNA polymerase, halting transcription. It is one of the most potent cytotoxics in use, dosed in micrograms per kilogram rather than milligrams per square metre.

Its indications are largely paediatric and rare: Wilms tumour, rhabdomyosarcoma, Ewing sarcoma, and gestational trophoblastic neoplasia, where single-agent therapy can be curative. It is given by intravenous injection. Severe myelosuppression and mucositis are expected. Extravasation causes progressive tissue necrosis and is a surgical emergency. Veno-occlusive disease of the liver is a specific risk in children, and the drug markedly enhances radiation injury, so concurrent radiotherapy requires dose adjustment.
