Product Description:

Copanlisib inhibits phosphoinositide 3-kinase, with activity concentrated on the alpha and delta isoforms. The delta isoform drives survival signalling in malignant B cells; the alpha isoform contributes an antiangiogenic effect. What distinguishes it from the oral drugs in the same class is the route: it is given as a short intravenous infusion on an intermittent schedule, which produces high peak exposure with drug-free intervals and was intended to avoid the cumulative immune toxicity that limits continuous oral dosing.

It is used for relapsed follicular lymphoma in adults after at least two prior systemic therapies. Because PI3K-alpha inhibition interferes with insulin signalling, transient hyperglycaemia occurs in most patients within hours of infusion and must be anticipated in diabetics. Hypertension follows the same pattern. Other risks include infusion-related reactions, non-infectious pneumonitis, severe neutropenia and opportunistic infection, with Pneumocystis prophylaxis recommended.
