Chlorzoxazone is a centrally acting skeletal muscle relaxant. It does not act on muscle itself; it works at the spinal cord and subcortical level, damping the polysynaptic reflexes that maintain painful muscle spasm. The practical effect is reduced spasm and better movement, usually alongside rest, physiotherapy and an analgesic.

It is used for short-term relief of discomfort from acute, painful musculoskeletal conditions. Its limiting problem is hepatotoxicity: idiosyncratic liver injury is rare but can be severe and unpredictable, and it is not dose-related, so patients are told to stop and seek advice if they develop nausea, dark urine or jaundice. Liver function is checked periodically with continued use. Drowsiness and dizziness are common, and it should not be combined with alcohol or other sedatives. A harmless orange or reddish discolouration of urine is expected and worth warning patients about so it is not mistaken for blood.

Sourcing notes. Off patent and inexpensive. It is registered mainly in the US and parts of Asia; in Europe it has largely been replaced by other muscle relaxants. Several strengths exist and prescribing differs by market, so buyers should confirm which strengths are registered locally.
