Promethazine injection is used to relieve or prevent some types of allergy or allergic reactions. It works by preventing the effects of a substance called histamine, which is produced by the body. Histamine can cause itching, sneezing, runny nose, and watery eyes.

Product Description:

Promethazine hydrochloride is a phenothiazine derivative acting as a potent first-generation H1 antagonist, with substantial anticholinergic, antiemetic and sedative activity. The injectable presentation is used for allergic reactions where the oral route is unsuitable, for nausea and vomiting, and as pre-operative sedation. Standard strengths are 25 mg and 50 mg per mL.

A serious administration constraint that must reach the end user. Promethazine injection carries a boxed warning in the United States concerning severe tissue injury, including gangrene, following intravenous administration. The drug is markedly caustic to tissue, and inadvertent intra-arterial injection or perivascular extravasation has resulted in tissue necrosis and amputation. Deep intramuscular injection is the preferred route; where intravenous administration is unavoidable, dilution and slow administration through a running line are specified.

It is also contraindicated in children under two years of age because of the risk of fatal respiratory depression.
