Product Description:
Mertansine, also called DM1, is a thiol-bearing derivative of maytansine that binds tubulin and stops cells dividing; it is far too toxic to be given alone and is used as the payload of antibody-drug conjugates. In trastuzumab emtansine, DM1 is attached to the antibody through the non-cleavable MCC linker, and that medicine is approved for HER2-positive metastatic breast cancer previously treated with trastuzumab and a taxane, and for HER2-positive early breast cancer with residual invasive disease after neoadjuvant treatment (reference brand: Kadcyla).

Regulatory files and timing
The FDA list of Drug Master Files (third quarter 2026) shows 5 active Type II API DMFs for this molecule. Few filings usually means few qualified sources, so early contact with suppliers pays off. For Europe, ask whether the supplier holds a CEP or an EU ASMF and, for imports into the EU, a written confirmation.
Generic or biosimilar entry. United States: No confirmed date (not documented). European Union: No confirmed date (not documented).
Handling and quality points
DM1 is among the most potent cytotoxics handled in pharmaceutical manufacture, so it needs isolator-level containment, dedicated equipment and occupational exposure limits well below one microgram per cubic metre. The free thiol oxidises readily to the disulfide dimer, so ask how the supplier prevents and limits this, and agree inert-atmosphere packaging and cold storage based on stability data. As the material goes into a conjugation step, buyers also look at maytansinoid-related impurities that could react and carry through into the final ADC.

Mertansine (DM1) API — third party manufacturing & bulk supply
Mertansine (DM1) API is available through PharmaTradz for wholesale and bulk supply as well as on a third party (contract) manufacturing basis. Buyers sourcing this product generally take one of three routes:
Indicative minimum order quantity
Mertansine (DM1) API is supplied as API (drug substance) — a dosage form priced by order value, so the indicative MOQ is a minimum order value of USD 5,000 per product. Actual MOQ is confirmed by the allocated manufacturer against your pack presentation and destination market. See full supply terms for dossier formats, market availability and shelf life.
Manufacturers of Mertansine (DM1) API listed on PharmaTradz hold site-level approvals across WHO-GMP, EU-GMP, USFDA, ANVISA and TGA, verified at onboarding.

Frequently Asked Questions
Expect a GMP certificate covering highly potent manufacture, the specification and certificate of analysis, a related-substances profile with identified maytansinoid impurities, and stability data. For commercial programmes, ask whether a US DMF exists for the payload or linker-payload, and request occupational exposure and toxicology data for your conjugation site.
It is semi-synthetic: a maytansinoid precursor, ansamitocin P-3, is produced by microbial fermentation and then converted chemically to DM1. Ask about fermentation controls, the purification steps and how related ansamitocins carried over from fermentation are limited.
Yes. PharmaTradz introduces ADC developers, CDMOs and research teams to GMP-compliant payload manufacturers with the containment this material needs, and we review their documents before any contact. Your project stays confidential until both sides agree to talk.
