Product Description:

Flupentixol is a thioxanthene antipsychotic, a first-generation drug that blocks dopamine D1 and D2 receptors. Its effect depends on dose: low doses have an antidepressant, mildly activating action, while higher doses are antipsychotic. This page covers tablets of 0.5 mg, 1 mg and 3 mg as the dihydrochloride. The 0.5 mg and 1 mg tablets serve the low-dose indication and the 3 mg tablet the psychosis indication. The decanoate depot injection is a separate product.

In several European markets low-dose tablets are licensed for depression, with or without anxiety, in adults. Treatment usually starts at 1 mg in the morning, with the last dose no later than mid-afternoon because the drug can be activating. Higher doses are used for schizophrenia and other psychoses. Pharmacists will expect the usual antipsychotic warnings: extrapyramidal symptoms, tardive dyskinesia, neuroleptic malignant syndrome and QT prolongation, plus higher mortality in older people with dementia. Other QT-prolonging drugs and alcohol need care.

Flupentixol is not approved in the US, but it is registered in the UK, much of Europe and various other markets, so check approval and the licensed indication for each strength in your destination. It is prescription-only. A CTD or ACTD dossier should include bioequivalence data, dissolution profiles for each strength and zone IVb stability for hot, humid climates. The pack, bottle or blister, should shield the coated tablets from light and moisture; storage is at room temperature.
