Product Description:

Clobazam is a 1,5-benzodiazepine. Like other benzodiazepines it strengthens the inhibitory effect of GABA at the GABA-A receptor, but its different ring structure gives a profile used mainly in epilepsy. Its active metabolite, N-desmethylclobazam, has a long half-life and carries much of the effect. Clobazam is given by mouth only; there is no injectable form in routine use, and the 5 mg, 10 mg and 20 mg strengths listed here are tablet strengths. Oral suspension and oral film exist in some markets for children.

Clobazam is widely used as add-on therapy when first-line antiepileptics have not controlled seizures. In the US it is approved for seizures of Lennox-Gastaut syndrome from two years of age, and in the UK it is also licensed for short-term relief of severe anxiety. Doses are built up gradually and tapered, never stopped abruptly, as withdrawal can provoke seizures. Sedation and dependence are the main concerns, and opioids or alcohol add dangerous respiratory depression. Poor CYP2C19 metabolisers may need lower doses.

Sourcing notes. Clobazam is a controlled medicine in many countries, usually listed with other benzodiazepines. Supply moves only when the importer holds the required import permit and the exporter the matching export authorisation, and quantities shipped must match those permits. It is prescription-only.
