Product Description:

This tablet combines low-dose acetylsalicylic acid (aspirin) 75 mg with magnesium hydroxide 15.2 mg. Aspirin irreversibly blocks platelet cyclooxygenase-1, stopping thromboxane A2 production for the platelet's seven to ten day lifespan and reducing clotting at doses far below those for pain. Magnesium hydroxide is a non-absorbable antacid included as a buffer, neutralising some acid around the dissolving tablet as an alternative to enteric coating. It does not remove the risk of gastrointestinal bleeding.

The tablet is used for long-term secondary prevention after myocardial infarction, ischaemic stroke or transient ischaemic attack, in angina and after coronary stenting or bypass. Primary prevention is now decided case by case because bleeding risk can outweigh benefit. One tablet is taken daily, often with food. Anticoagulants, other antiplatelets, NSAIDs, SSRIs and corticosteroids add to bleeding risk, and regular ibuprofen can blunt the antiplatelet effect. The magnesium can reduce absorption of tetracyclines, quinolones and iron. It is avoided under 16 because of Reye's syndrome.

Sourcing notes. This fixed combination is not registered everywhere, so check whether your regulator already lists it; if not, the CTD or ACTD dossier may need a justification for the combination. Aspirin hydrolyses to salicylic and acetic acid in moisture, so free salicylic acid is the key stability parameter. Ask for its limit and result on every batch CoA, plus Zone IVb data for hot, humid markets. Alu-alu blisters or sealed bottles with desiccant protect the tablets, and a vinegar smell signals degradation. Request GMP and CoPP documents.
