Product Description:
Acalabrutinib is a second-generation Bruton tyrosine kinase (BTK) inhibitor that binds covalently to the enzyme and shuts down the B-cell receptor signals that malignant B cells rely on. It is approved for adults with chronic lymphocytic leukaemia or small lymphocytic lymphoma, for mantle cell lymphoma after at least one prior therapy, and, with bendamustine and rituximab, for previously untreated mantle cell lymphoma in patients not eligible for stem cell transplant (reference brand: Calquence).

Regulatory files and timing
The FDA list of Drug Master Files (third quarter 2026) shows 7 active Type II API DMFs for this molecule. Few filings usually means few qualified sources, so early contact with suppliers pays off. For Europe, ask whether the supplier holds a CEP or an EU ASMF and, for imports into the EU, a written confirmation.
Generic or biosimilar entry. United States: Patent expiry; entry not confirmed (Nov 2026 (earliest DS patent)). European Union: No confirmed date.
Handling and quality points
Be clear from the start which drug substance your formulation needs, the free base or the maleate monohydrate, and make sure the supplier controls that solid form; for the maleate, hydrate state and salt stoichiometry need checking batch to batch. As a covalent kinase inhibitor with a reactive butynamide group, acalabrutinib is normally handled under high-potency controls, and buyers should ask how reactive and potentially mutagenic impurities are controlled. A nitrosamine risk assessment should form part of the standard package.

Acalabrutinib API — third party manufacturing & bulk supply
Acalabrutinib API is available through PharmaTradz for wholesale and bulk supply as well as on a third party (contract) manufacturing basis. Buyers sourcing this product generally take one of three routes:
Indicative minimum order quantity
Acalabrutinib API is supplied as API (drug substance) — a dosage form priced by order value, so the indicative MOQ is a minimum order value of USD 5,000 per product. Actual MOQ is confirmed by the allocated manufacturer against your pack presentation and destination market. See full supply terms for dossier formats, market availability and shelf life.
Manufacturers of Acalabrutinib API listed on PharmaTradz hold site-level approvals across WHO-GMP, EU-GMP, USFDA, ANVISA and TGA, verified at onboarding.

Frequently Asked Questions
Ask for a GMP certificate, the specification and certificate of analysis for the exact form you need, and a US DMF letter of access or EU ASMF where they exist. Stability data, solid-form characterisation, a mutagenic impurity assessment and a nitrosamine risk assessment round out the usual set.
That depends on the product you are developing. The capsules were approved with the free base, while the tablets use the maleate monohydrate, which differs in solid-state and handling behaviour. Your bioequivalence plan and dossier must match the form, so settle this before you ask for samples.
Yes. PharmaTradz brings licensed manufacturers, CDMOs and R&D teams together with GMP-compliant kinase inhibitor producers, reviewing documents before any introduction. Buyer details are kept private until both sides agree.
