Product Description:
Vorasidenib blocks mutant forms of the enzymes IDH1 and IDH2. In IDH-mutant gliomas these enzymes produce 2-hydroxyglutarate, a metabolite that disturbs how genes are switched on and drives tumour growth. Vorasidenib crosses into the brain and lowers 2-hydroxyglutarate in the tumour. It is the first targeted therapy for these gliomas.
The US FDA approved vorasidenib as Voranigo in August 2024, and the European Commission followed in September 2025. It is used for grade 2 astrocytoma or oligodendroglioma with a susceptible IDH1 or IDH2 mutation, in patients aged 12 and over, after surgery. In its main trial it delayed progression and the need for radiotherapy and chemotherapy. It is taken once a day, usually 40 mg (20 mg for patients under 40 kg), and liver tests are monitored.
