Regulatory files and timing

The FDA list of Drug Master Files (third quarter 2026) shows 1 active Type II API DMF for this molecule. Few filings usually means few qualified sources, so early contact with suppliers pays off. For Europe, ask whether the supplier holds a CEP or an EU ASMF and, for imports into the EU, a written confirmation.

Generic or biosimilar entry. United States: Patent expiry; entry not confirmed (2033). European Union: Protected beyond 2033 (2034).

Handling and quality points

Lorlatinib is normally handled as a high-potency kinase inhibitor, so contained weighing and dispensing, and a clear exposure band from the maker, are standard requests. It is a single enantiomer, so chiral purity belongs in the specification alongside the crystalline form. Aqueous solubility falls steeply as pH rises (from about 32 mg/mL at pH 2.55 to 0.17 mg/mL at pH 8.02), which makes particle size worth agreeing early; ask too for the nitrosamine risk assessment.

Lorlatinib API — third party manufacturing & bulk supply

Lorlatinib API is available through PharmaTradz for wholesale and bulk supply as well as on a third party (contract) manufacturing basis. Buyers sourcing this product generally take one of three routes:

Wholesale / bulk supply Stock or scheduled supply from an existing manufacturer. The fastest route where the product is already registered in your market.
Third party manufacturing Manufactured to your specification and packed under your own brand and label — suits distributors and brand owners building a portfolio in their market.
Dossier licensing Where a CTD, EU-CTD or ACTD dossier exists it can often be licensed for filing in your name, compressing market entry from years to months.

Indicative minimum order quantity

Lorlatinib API is supplied as API (drug substance) — a dosage form priced by order value, so the indicative MOQ is a minimum order value of USD 5,000 per product. Actual MOQ is confirmed by the allocated manufacturer against your pack presentation and destination market. See full supply terms for dossier formats, market availability and shelf life.

Manufacturers of Lorlatinib API listed on PharmaTradz hold site-level approvals across WHO-GMP, EU-GMP, USFDA, ANVISA and TGA, verified at onboarding.

Frequently Asked Questions

A GMP certificate for the site, the specification with a recent CoA, and either a US DMF letter of access or an EU ASMF for the filing. Most teams also want long-term and accelerated stability data, the impurity and nitrosamine assessments, and containment information for this potent compound.

Lorlatinib has been described in more than one crystalline form, including solvates, and form patents can affect generic launch timing. Agree the form with your supplier and patent advisers, ask for XRPD data against your reference and check the form stays stable under your process conditions.

We can. PharmaTradz links licensed manufacturers, CDMOs and R&D groups with GMP-compliant lorlatinib makers, and we look at the documents before putting anyone in touch. Neither side's details are shared until both agree.
