Concizumab is a monoclonal antibody used for the routine prophylaxis of bleeding episodes in patients with hemophilia A or B, with or without inhibitors. It works by inhibiting tissue factor pathway inhibitor (TFPI), helping restore blood clotting and reduce the frequency of bleeds. The drug is administered as a subcutaneous injection using a prefilled pen. It is indicated for adolescents and adults aged 12 years and older.

Concizumab is a monoclonal antibody against tissue factor pathway inhibitor, working on the same principle as other agents in this class: rather than supplying a missing clotting factor, it disables one of the body's natural restraints on coagulation, allowing enough thrombin to be generated for a clot to form. Because it acts below the point where factors VIII and IX operate, it is effective across all four haemophilia populations — A and B, with and without inhibitors.

It is used for routine prophylaxis of bleeding episodes in haemophilia A or B with inhibitors, and in some markets without, given by daily subcutaneous injection from a prefilled pen after a loading dose. Daily dosing is more frequent than some alternatives, but the coverage across inhibitor status is broader. Injection site reactions are common. As with the class, thromboembolic events are the risk that defines the dosing strategy, and the development programme was paused and restarted over exactly this issue.

Sourcing notes. Originator biologic, on patent, recombinant rather than plasma-derived. Three pen strengths cover loading and maintenance and all are needed for a treatment course. Cold chain at 2–8 degrees, with a limited in-use period at room temperature once started. Registration varies by market; confirm before quoting.
